摘    要:目的:改良Golgi-Cox染色法,使操作时间缩短、染色恒定;探讨该方法在脑缺血研究中的应用价值。方法:通过多聚甲醛固定组织、缩短浸胶时间、组织块外包石蜡壳等方法,比较研究了改良法与传统方法在显示神经元数量、操作时间等方面的差别;比较研究了大鼠脑缺血1h再灌注2、4、8和12周缺血侧神经元数量等的动态变化。结果:改良后使操作时间缩短为5-6h;染色神经元数量明显多于传统方法;脑缺知后缺血区神经元数量呈进行性减少,缺血周围残存的神经元突起增加。结论:改良Golgi-Cox神经元染色法较恒定,且省时,在脑缺血后神经元可塑性研究中有较大的应用价值。

Abstract:Objective:To improve the method of Golgi-Cox staining and get it more convenient in the studing of pathogenesis of ischemia in rats.Methods:In the improved group ,the rat brains were fixated by 4% paraformaldelhyde fixative in 1,2,4,12 weeks after one hour of middle cerebral artery occlusion,then immerse the brain in 5% collodion solution for 60 -120 min. A rind of paraffin was made outdise the blok.The neurons in the field of ischemia were numbered.Reults:After improving,the procedure was decreased to 5-6 hours.Moreover, using this method , the number of stained neurons was stable in the rat brain.It was decreased gradually after ischemia,but the damaged neuron had more dendrites in the perilesion.Conclusion:The method is not only saving time,but also more stable than the one reported before.It may be used in the studing of plasticity after ischemia.
