菟丝子多糖对衰老小鼠脂质过氧化的影响

摘    要:目的:观察菟丝子多糖对衰老小鼠模型丙二醛(MDA)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSHPX)和脑组织中脂褐质(LF)的影响。方法:用d半乳糖制备衰老小鼠模型,随机分为模型组(不给药)、阳性对照组(灌注维生素E200mg·kg-1,qd)、3个剂量菟丝子多糖组(分别灌注100,200,400mg·kg-1菟丝子多糖,qd),共7wk。另设空白对照组。结果:模型小鼠血清、肝、肾中MDA含量升高,SOD及GSHPX活性降低,脑中LF升高。而100,200,400mg·kg-1·d-1菟丝子多糖组能不同程度对抗模型小鼠脑组织中LF升高及血清、肝、肾组织中MDA升高,其量效关系均呈负相关,r分别为-0.916,-0.939,-0.950及-0.911,P<0.01;能不同程度抑制血清、肝、肾组织中SOD活力下降,其量效关系均呈正相关,r分别为0.898,0.939及0.946,P<0.01;能不同程度抑制血清、肝、肾组织中GSHPX活力下降,其量效关系均呈正相关,γ分别为0.947,0.969及0.947,P<0.01。结论:菟丝子多糖有显著清除氧自由基及抗脂质过氧化作用。

Abstract:To observe the effects of polysaccharide from Cuscuta chine nsis (PCC)on malondialdehyde (MDA), superoxide dismutase (SOD) and glutat hi one peroxidase (GSH-PX) in serum, liver, kidney and lipofuscin(LF) in brain of senile model mice. METHODS: The senile model mice caused by d -galatose (d-gal), were randomly divided into model group (no administer d rug), positive control group (gavage Vit E 200 mg·kg~(-1),qd), three differe nt doses of PCC groups (gavage PCC 100,200,400 mg·kg~(-1),qd, respectively ) for 7 wk. Another blank group was controled. RESULTS:The model group showed elevations of MDA in serum, liver, kidney and LF in brain; conversely with descent of SOD and GSH-PX activities. The three diff erent doses of PCC groups demonstrated different degrees of antagonistic negativ e correlation ship for dosage effects on elevations of cerebral LF, and the MDA in serum, liver, kidney of model mice, with -0.916,(-0.939),(-0.950) and (-0.911) r, respec tively, P<0.01; furthermore revealid various inhibitions on descend activiti es of SOD and GSH-PX in serum, liver, kidney with dosage effects of positive co rrelationship 0.898, 0.939 and(0.946) r,P<(0.01); and (0.947),( 0.969), (0.947)[JP] r,(P<0.01), respectively. CONCLUSION: PCC possesses the notable effects of eliminating oxyen free radicals and antilipoperoxidation.
