Toll样受体4在支气管哮喘患者气道平滑肌中的表达研究

目的 探讨支气管哮喘(简称哮喘)患者气道平滑肌上Toll样受体4(TLR4)、增殖细胞核抗原(PCNA)及B淋巴细胞瘤-2基因(Bcl-2)蛋白表达与气道平滑肌细胞的增殖、凋亡及气道重塑的关系.方法 收集2014年1月至2015年12月,哮喘患者8例肺组织及非哮喘患者8例肺组织,分别作为实验组及对照组;测量2组气道壁厚度以对比其差异;并用免疫组织化学法检测TLR4、PCNA、Bcl-2在实验组及对照组气道平滑肌上的表达差异.结果 实验组与对照组气道壁厚度测定有显著差异[(80.91±4.99) μmvs (50.39±4.51) μm,t=12.858,P＜0.001],实验组气道平滑肌细胞上TLR4、PCNA、Bcl-2表达明显升高(137.82±7.79 vs 203.79±5.94,t=19.045,P＜0.001;119.72±7.50 vs211.87±9.43,t=21.634,P＜0.001;109.54±11.34vs204.53±9.03,t=18.533,P＜0.001).结论 哮喘患者气道平滑肌中TLR4、PCNA、Bcl-2的过表达与气道平滑肌细胞增殖增加、凋亡抑制及气道重塑有关.

Study of Toll-like receptors 4 expression in airway smooth muscle of asthmatics

Objective To study the relationship between the protein expression of Toll-like receptor 4 (TLR4),proliferating cell nuclear antigen (PCNA) and Bcl-2 in the airway smooth muscles of asthmatics and airway smooth muscle cell proliferation,apoptosis and airway reconstruction.Methods Eight cases of lung tissue in asthmatics and 8 cases of lung tissue in patients not with asthma were collected from 2014 to 2015,respectively as the experimental group and control group.The ipsilateral airway wall thickness of the two groups were measured and the differences were compared.TLR4,PCNA,and Bcl-2 expression in the airway smooth muscle of the two groups were detected by immunohistochemistry.Results There are significant differences in the airway wall thickness between the experimental group and control group [(80.91 ± 4.99) μm vs (50.39 ± 4.51) μm,t =12.858,P ＜ 0.001].TLR4,PCNA and Bcl-2 expression in airway smooth muscle cells were significantly higher in the experimental group than in the control group (137.82±7.79 vs 203.79±5.94,t =19.045,P ＜0.001;119.72±7.50 vs211.87±9.43,t=21.634,P＜0.001;109.54±11.34vs204.53±9.03,t=18.533,P＜ 0.001).Conclusions There are significant correlations between the overexpression of TLR4,PCNA and Bcl-2 in the airway smooth muscles of asthmatics and airway smooth muscle cell proliferation,apoptosis suppression and airway reconstruction.
