摘    要:目的探讨骨桥蛋白(OPN)相关基因在口腔癌侵袭转移中的作用。方法采用免疫组织化学SP法,检测OPN、核因子-κB p65(NF-κB p65)和基质金属蛋白酶-9(MMP-9)在58例口腔癌和10例正常口腔粘膜中的表达。结果在10例正常口腔粘膜组织中,OPN、NF-κB p65和MMP-9均无阳性表达。在32例无颈淋巴结转移的口腔癌组中OPN、NF-κB p65和MMP-9的表达率依次为40.6%、34.4%和37.5%,在26例伴有颈淋巴结转移的口腔癌组中其阳性率依次为69.2%、65.4%和84.6%,其阳性表达率在有转移的口腔癌组中高于无转移的口腔癌组,两组比较差异有统计学意义(P〈0.05),而与患者性别、年龄、肿瘤大小、组织学分级均无相关性(P〉0.05)。OPN和NF-κB p65的表达与NF-κB p65和MMP-9的表达均呈正相关(r1=0.57,r2=0.52,P〈0.05)。结论 OPN可能通过活化NF-κB,上调转移相关分子MMP-9的表达,从而促进口腔癌细胞的侵袭转移。

The expressions of osteopontin and its related genes in oral squamous cell carcinoma

Abstract:Objective To investigate the correlation between the expression of osteopontin(OPN) related genes and invasion and metastasis in oral squamous cell carcinoma(OSCC).Methods The expression of OPN,nuclear factor-κB p65(NF-κB p65) and matrix metalloproteinase 9(MMP-9) was detected by immunohistochemistry in 10 cases of normal oral mucosa and 58 cases of OSCC.Results OPN,NF-κB p65 and MMP-9 were not expressed in normal oral mucosa.The expression rate was 40.6%,34.4% and 37.5% respectively in 32 OSCC samples without cervical lymph node metastasis,but the rate increased to 69.2%,65.4% and 84.6% respectively in 26 OSCC samples with cervical lymph node metastases.There was significant difference in the expression rate between the group with or without cervical lymph node metastasis(P0.05),there was no significant difference between the expression rate and gender,age at diagnosis,tumor size and histological grade(P0.05).Correlations existed between OPN and NF-κB p65 as well as between NF-κB p65 and MMP-9(r1=0.57,r2=0.52,P0.05).Conclusion Osteopontin overexpression is closely related to the invasion and metastasis of OSCC through MMP-9 up-regulation via activating NF-κB p65.
