祛痰通络汤对早期糖尿病肾病大鼠足细胞nephrin蛋白表达的影响

目的 观察祛痰通络汤对糖尿病肾病大鼠足细胞裂孔隔膜蛋白分子(nephrin)表达的影响,探讨其防治DN的作用机制.方法 制作大鼠糖尿病肾病模型,分为正常对照组、模型组、贝那普利治疗组及祛痰通络汤低、中、高剂量组,干预8周.观察各组24h尿蛋白定量、肌酐、肾脏病理学变化;免疫组化观察肾组织nephrin蛋白表达的变化.结果 经过祛痰通络汤治疗后,24h尿蛋白定量下降;病理改变减轻;nephrin蛋白表达增加.结论 祛痰通络汤对DN大鼠具有一定的肾脏保护作用,其机制可能与恢复肾组织足细胞nephrin蛋白表达有关.

Effect of Qutantongluo Decoction on Protein Expression of Nephrin in Podocytes of Early Diabetic Nephropathy Rats

Objective:To observe the effects of Qutantongluo Decoction on the expression of nephrin in podocytes of diabetic nephropathy rats.Methods:Rats models of diabetic nephropathy were established by injection with streptozotocin intraperi-toneally.Wistar rats were randomly divided into 6 groups: the normal group,the DN group,the benazepril hydrochloridec group,the Qutantongluo Decoction low dose group,the Qutantongluo Decoctione median dose group and the Qutantongluo Decoction high dose group.All rats were gavaged once daily for 8 weeks.24 h urinary protein and creatinine were determined.Renal tissue were also analyzed by pathological examination.Results:The protein expression of nephrin was detected by immunohistochemistry.After Qutantongluo Decoction intervention,levels including the 24h total urine protein and the SCr were significantly reduced than those in STZ group.The renal pathological status in Qutantongluo Decoction groups were significantly improved and the protein expression levels of nephrin also were markedly increased.Conclusion:The kidney injury in the rats with diabetic nephropathy is ameliorated by Qutantongluo Decoction,which may be related to restoration of the protein of nephrin.
