中药熏洗对大鼠失神经支配骨骼肌NGF与bFGF影响的实验研究

摘    要:目的观察中药熏洗一号方熏洗对大鼠失神经支配骨骼肌神经生长因子(NGF)与碱性成纤维生长因子(bFGF)影响。方法雄性SD大鼠120只,随机取90只造模,成功后随机分为熏洗一号方组、丹参组、模型组各30只,另30只作空白对照组。分别在造模后第7天、14天和21天各处死10只,股直肌肌腹组织块取材,观察失神经支配后骨骼肌结构与NGF、bFGF免疫组织化学染色。结果(1)NGF表达:术后第7天:熏洗一号方组,NGF表达明显成于其它各组;术后第14天:熏洗一号方组,NGF表达明显成于其它各组,丹参组NGF表达显示;术后第21天:熏洗一号方组,丹参组纤维组织未见染色。(2)bFGF表达:术后第7天:熏洗一号方组和丹参组成纤维细胞染色阳性,模型组高达阳性。术后第14天:熏洗一号方组、丹参组表达明显,术后第21天:熏洗一号方组表达明显优于其它各组。结论失神经支配骨骼肌损伤的修复与NGF和bFGF的变化有关,骨骼肌失去神经生长因子(NGF)的营养发生萎缩,bFGF能促进运动神经纤维的再生。

Abstract:Objective : To study the mechanism of Xunxiyihaofang on the recovery of NGF and bFGF of denervated skeletal muscle. Method: 120 Male SD rats were selected, 90 were randomly divided into three groups -Xunxiyihaofang group, Salvia miltiorrhiza group and model group. And another 30 rats were in control group. 10 of experimental animals in each group after 7 days, 14 days and 21 days were killed, to draw the materials from rectus femoris venter musculi tissue, conventional HE staining, to observe ultrastructural of denervated skeletal muscleuhramicrostructure with transmission electron microscopy after changes and NGF and bFGF immunohistochemical staining. Results: ( 1 ) The expression of NGF: After seven days, Xunxiyihaofang group, muscle spindle myocyte staining positive, tibroblasts, vascular end Othelial Ceils were weak, Salvia miltiorrhiza group muscle Spindle myocyte were weakly positive. Model group muscle Spindle smaller, there was no obvipusstaining. After 14 days, Xunxiyaofang group, myocyte cellul arnucleu is not smooth, Staning positive, fibroblast proliferation and staining positive strongly, vascularendOthelial cellS positive staining, muscle Spindle no obvious staining, Salviamihiorrhiza group myocyte cellular nucle us invagination, positive staining and fibroblasts positive staining. Model group and control group muscle spindle atrophy, negative staining. After 21 days, Xunxiyihaofang group, staining thin, endocytoplasmic reticulum exvansion and weak positive, Salvia miltorrhiza group myocyte cytolemma shrinkage and no staining. (2) expression of bFGF: After seven days, Xunxiyihaofang group myocyte staining positive, Salvia miltiorrhiza group and model group fibroblastpositive staining.
