摘    要:
目的构建人肠三叶因子(hITF)的毕赤酵母表达载体。方法从人结肠黏膜提取总RNA,经RT-PCR得到hITF cDNA,用PCR方法扩增hITF基因,并将其克隆到毕赤酵母表达载体pPIC9中,构建含AOX1启动子和α分泌信号肽序列的重组表达质粒pPIC9/hITF。结果通过酶切和基因序列分析确定插入pPIC9中的片段为hITF基因片段。结论重组质粒pPIC9/hITF的成功构建,为在真核细胞中高效表达hITF。并制备其抗体的进一步研究提供基础。

Abstract:
Purpose To construct Pichia pastoris expression vector of human intestinal trefoil factor. Methods Total RNA was extracted from human colon mucosa, hITF cDNA was isolated by RT-PCR, hITF gene was amplified with PCR and cloned into Pichia pastoris vector pPIC9. The expression plasmid pPICg/hlTF containing AOX1 promotor and the sequences of asecreting signal peptides was constructed. Results The nucleotide sequence of hITF was the same as expected. Conclusion The successful construction of the recombinant plasmid will be further studied in this field.
