摘    要:为了快速观察组织工程心脏瓣膜构建中种植细胞的生长状况,采用去细胞猪主动脉瓣叶为支架,将新生牛主动脉内皮细胞种植于其上,第3、7天取瓣叶行H-E直接染色,并以常规切片H-E染色和扫描电镜作为对照。结果发现,该3种方法观察所见细胞第3、7天的生长疏密状况基本同步。提示,瓣叶H-E直接染色可方便快捷地观察种植细胞的生长状况。

Abstract:To rapidly observe the growth of cells seeded on the scaffold during tissue engineering of heart valve, acellularized porcine aortic valve leaflets were used as scaffold, then bovine aortic endothelial cells were seeded on the scaffold. At day 3 and 7 after cells seeded, recellularized leaflets were stained directly with H E, and histological sections were also stained with H E,scanning electron microscopy of recellularized leaflets were performed as control. The results showed that 3 methods were able to observe the growth of the cells seeded on the scaffold. At day 3 and 7 after cells seeded, cell distributions on the scaffold were similar. Valve leaflets stained directly with H E can observe the growth of cells rapidly and conveniently.
