糖心康对糖尿病大鼠心肌细胞凋亡促进与抑制因子基因表达的调控作用

摘    要:目的探讨糖心康对糖尿病心肌损伤的保护作用及机理。方法采用STZ加高脂饮食造模。治疗组以中药糖心康灌胃,空白组及模型组以生理盐水灌胃,以基因芯片技术TUNEL法检测心肌细胞凋亡。结果模型组细胞凋亡较正常组发生率明显增加,两组比较差异非常显著(P<0.01),而糖心康组心肌凋亡率则下降,模型组大鼠Fas基因蛋白表达增强,与正常组比较差异非常显著(P<0.01),而糖心康组Fas基因表达减弱,与模型组比较差异显著(P<0.01),模型组Bcl-2表达减弱,而糖心康组Bcl-2表达较模型组增强,二者比较差异显著(P<0.01)。利用基因芯片技术在心肌基因表达谱中筛选出与细胞凋亡有关的促进与抑制因子基因主要有2个,即AFO41376,M75720。结论糖心康有抑制心肌细胞凋亡作用,可使Fas基因、AFD41376基因表达下调,使Bcl-2,M75720表达上调。这也是中药复方糖心康具有心肌损伤保护作用的机制之一。

Abstract:Objective To discuss the mechanism and protective effect of Tang-Xinkang against myocardial damage of diabetic mellitus rats.Methods The myocardial damage models of diabetic mellitus rats were created by STZ and high calorie feed.Treat- ment group rats were administered Tangxinkang while the control group and model group were given physiological saline.TUNEL method and gene chip technology were applied to examine myocardial apoptosis.Results The occurrence rate of apoptofie cell in- creased more in the model group than the control group(P<0.01),apoptotic cell was lower in the treatment group,while the ex- pression of Fas was stronger in the model group.The difference was significant compared with the control group(P<0.01).The expression of Fas was lower in the treatment group,the difference was significant compared with the model group(P<0.01).The expression of Bcl-2 was lower in the model group,while the expression was stronger in the treatment group,the difference was significant between them(P<0.01).Through adopting gene chip technology,we found two mainly promotive and inhibitory genes related to apoptosis in the Gene expression profiling,those were AF041376 and M75720 genes.Conclusion Tang Xin-kang can restrain myocardial apoptosis,down-regulate the expression of Fas and AF041376,while up-regulate Bcl-2 and M75720.It is one of protective mechanisms of the Chinese compound.
