Th17细胞及相关细胞因子在原发性胆汁性肝硬化患者的表达

摘    要:目的观察PBC患者外周血Th17/CD4+T细胞相关细胞因子(IL-17、IL23、IL21、IL6、TGFβ)的表达,探讨Th17/CD4+T细胞在PBC发病中的作用.方法收集26例PBC患者(PBC组),20例HBV患者(HBV组)和20例健康人群(HC组)的外周血,采用酶联免疫吸附法(enzyme-linked immunosorbent assay,ELISA)测定3组患者血清IL-17、IL-23、IL-21、IL-6、TGFβ的水平,比较3组间的表达情况.结果 PBC患者血清IL-17、IL-23、IL-21的表达明显高于HBV患者和健康对照组(P〈0.05);PBC组IL-6、TGFβ表达明显高于健康对照组(P〈0.05),而与HBV组比较差异无统计学意义(P〉0.05).结论 Th17/CD4+T细胞及其分泌的特异细胞因子在PBC的免疫发病发挥着重要作用.

Abstract:Objective To observe the serum profile of Th17-related cytokines(IL-17, IL23, IL21, IL6 and TGFβ) in patients with primary biliary cirrhosis, so as to investigate the role of Th17 cells in the pathogenesis of primary biliary cirrhosis(PBC). Methods Peripheral blood samples of 26 PBC patients,20 HBV infected patients and 20 healthy subjects were collected. Serum levels of the abovementioned five Th17-related cytokines were determined by enzyme linked immunosorbent assay(ELISA). Results The serum levels of IL-17,IL23 and IL21 in PBC group were significantly higher than those in HBV group or HC group(P0.05);the serum levels of IL6 and TGF-β were comparable between the PBC group and HBV group, while the serum levels of IL6 and TGF-β were significantly higher in the PBC group than those in the HC group(P0.05). Conclusion Th17 cells and its related cytokines may play an important role in the development and progression of PBC.
