酶联免疫斑点方法改进及应用

摘    要:建立了ELISPOT方法并通过改进 ,应用到抗人CD8杂交瘤细胞及LACA小鼠体外脾细胞分泌IgG量的检测 ,同时观察六味地黄多糖CA4 3在体外对LACA小鼠脾细胞分泌IgG的影响。结果表明 10 μg/mlLPS作用下可明显提高小鼠脾细胞体外产生抗体的量和提高分泌抗体的细胞数。 5 0 μg/mlCA4 3在体外与LPS具有相似的作用 ,可以刺激小鼠脾细胞分泌IgG的量和分泌抗体的细胞数量增多。

Abstract:A modified enzyme-linked immunospot (ELISPOT) method was established and had been used for the detection of mouse IgG secretion in the anti-human CD8 hybridoma cells and the LACA mouse spleen cells. This method had been utilized to study the effect of "Liu Wei Di Huang" polysaccharide CA4-3 on the IgG secretion of of mouse splenocytes in vitro. The results showed that 10 μg/ml of LPS could enhance the in vitro production of antibodies produced by mouse splenocytes and increase the number of antibody secreting cells,meanwhile,50 μg/ml of CA4-3 had the similar effect as that induced by LPS.
