摘    要:目的 探讨胆固缺乏时对Jurkat细胞增殖功能及凋亡的影响。方法 用Lovastatin抑制Jurkat细胞内源性胆固醇的合成,以低浓度脂蛋白受体(LDL-R)介导的低密谋脂蛋白(LDL)内吞提供细胞外源性胆固醇,细胞处理1~3天后,用流式细胞仪分析各细胞周期相绫分布,并定量分析胆固醇缺乏对处理3天后细胞凋亡的影响。结果 Jurkat经去脂血清培养基加Lovastatin处理3天后,细胞受阻于Cu

Abstract:Objective To investigate the effects of cholesterol deficiency on the proliferation and apoptosis of Jurkat cells. Mothods Lovastatin was used to inhibit the endogenous synthesis of cholesterol of Jurkat cells. Exogenous cholesterol was provided with the the intake of low density lipoprotein (LDL) through the mediation of LDL-receptor (LDL-R). The cycle distribution and apoptosis of Jurkat cells were observed with flow cytometry (FCM) after the cells were cultured with different media for 1 to 3 days. Results After the administration of lovastatin for 3 days, the proliferation of Jurkat cells was inhibited and the cell cycle was blocked at the G0/G1 phase with the appearance of apoptosis. After the intake of LDL, a partial cessation of above mentioned changes was found. Conclusion Cholesterol deficiency induced with either inhibiting endogenous synthesis or decreased exogenous intake can result in the inhibition of proliferation or even apoptosis of Jurkat cells. which is positively correlated to the severity of cholesterol deficiency.
