| 摘 要： | 目的研究天花粉蛋白(TCS)对人宫颈癌HeLa细胞生长的抑制作用,探讨TCS抗肿瘤的作用机制。方法采用MTT法检测TCS对Hela细胞的抑制作用,形态学观察、DNA电泳及流式细胞仪检测TCS对HeLa细胞的诱导凋亡作用。结果TCS对HeLa细胞的生长具有明显的抑制作用,显微镜下可见细胞圆缩,胞浆凝聚等凋亡细胞的形态学改变,DNA凝胶电泳呈梯级格局,流式检测出现凋亡峰,凋亡比例具有剂量和时间依赖性。结论天花粉蛋白通过诱导细胞发生凋亡而抑制宫颈癌HeLa细胞的增殖。

| Abstract: | Objective To study the inhibitory effect of trichosanthin(Tcs) on Hela cells and its anti-tumor mechanism.Methods The antiproliferative effect of TCS on Hela cells was measured with MTT assay.Light microscopy was used to observe the morphology changes,DNA agarose electrophoresis and FCM analysis were carried out to examine the induced apoptosis.Results The proliferation of HeLa cells was significantly inhibited by TCS,Hela cells showed a serial of characteristic changes of apoptosis such as cell volume shrinkage,chromatin condensation;the occurrence of DNA ladder and apoptosis rate of Hela cells was dose and time dependent.Conclusion Trichosanthin can inhibit the proliferation of Hela cells by its induction of apoptosis. |
