非压迫性腰椎间盘髓核突出模型的建立和神经根组织学观察

摘    要:目的 设计一种新的非压迫性腰椎间盘髓核突出动物模型,观察神经根组织形态学变化。方法 16只sprague-Dawley(SD)雄性大鼠随机分成对照组和实验组,分别将自体肌肉混悬液和尾椎髓核混悬液注射到腰椎硬膜外腔,对其机械刺激缩爪阈值和神经根组织形态学进行观察。结果在无机械压迫的情况下,大鼠硬膜外髓核可产生明显的痛觉过敏反应,神经根组织形态产生明显改变。结论 大鼠硬膜外自体髓核注射法建立的非压迫性腰椎间盘髓核突出动物模型有效、可靠、费用低廉,是研究腰椎间盘突出症和根性神经痛较理想的动物模型。

Abstract:Objectives To design a new non-compressive animal model of lumbar nucleus pulposus prolapse and histologic changes of rat cauda equina nerve roots were observed.Methods 16 SD(Sprague-dawley)rats were chosed. The rats were divided into control group and experimental group . The suspension of muscle or nucleus pulposus of coccygeal vertebral was injected into the epidurale cavum of rat, making the non-compressive model of experimental lumbar nucleus pulposus prolapse. The histologic changes of rat cauda equina nerve roots and the mechanical withdraw threshold were observed.Results Without mechanical compression, rats autologous nucleus pulposus displayed significant hyperalgesia and histologic changes of rat cauda equina nerve roots was pronounced. Conclusions The non-compressive animal model offered to study the mechanism of disc herniation and nerve root pain is effective , reliable and cheap.
