表皮肿瘤中COX-2与P53的检测

目的了解表皮肿瘤组织环氧合酶-2(COX-2)与P53的表达情况。方法免疫组化法检测脂溢性角化病(SK)15例、Bowen’s病(BD)15例、基底细胞上皮瘤(BCE)20例、鳞癌(SCC)20例COX-2和P53的表达。结果所检测的各肿瘤组织标本均有COX-2的表达,分别为SCC95%,BD73.3%,SK46.6%,BCE68.0%,表达强度以SCC最为显著,周围正常组织未见表达。突变型P53在SCC(80.0%),BCE(75.0%),BD(33.3%)肿瘤组织的阳性率较高,而在SK(13.3%)中基本不表达。SCC和BCECOX-2表达阳性者其突变型P53表达的阳性率较COX-2表达阴性者高(P<0.05)。结论表皮肿瘤存在COX-2的过表达;P53的突变与COX-2的过表达有关,p53突变可能是通过上调COX-2水平发挥抗凋亡作用,从而促进SCC,BCE的形成和发展。

Detection of COX-2 and P53 in Skin Neoplasms

Objective To investigate COX-2 expression and the mutation of P53 in skin neoplasms.Methods COX-2 and P53 were detected by immunohistochemical staining in 15 seborrheic keratosis(SK),15 Bowen’s disease(BD),20 basal cell epithelioma(BCE),20 squamous-cell carcinoma(SCC) tissues.Results COX-2 expression could be observed in these 4 kinds of skin neoplasms,but not the normal tissues surrounding it.It was SCC 95%,BD 73.3%,SK 46.6%,BCE 68.0% respectively and stronger in SCC.Mutated P53 expression could be observed in SCC(80.0%),BCE(75.0%),BD(33.3%),but not normal tissue and SK(13.3%).Expression of mutated P53 was significant higher in the group of positive COX-2 staining than that in negative group in SCC and BCE(P<0.05).Conclusion These results indicate that COX-2 is up-regulated in skin neoplasm.The mutation of P53 is correlated with the over-expression of COX-2 protein.The P53 mutation may act as a anti-apoptosis factor in the development of SCC and BCE by up-regularated the expression of COX-2.
