人羊膜负载猪角朊细胞重建表皮的形态学研究

目的 采用细胞组织工程技术，将人羊膜（Human amniotic membrane,HAM）负载培养猪角朊细胞构建皮肤表皮替代物，并观察猪角朊细胞在HAM上生长增殖的形态特点。方法 将贵州小香猪的角朊细胞原代培养，传代扩增后，接种于HAM基质面，逐日于倒置显微镜下观察角朊细胞的生长增殖变化情况，并于培养3－15d进行光镜、电镜观察，检测角朊细胞在HAM上的生长情况。结果 接种后30min内就明显见到角朊细胞在HAM上贴附，24h内大部分贴附生长，3d形成单层完全覆盖HAM。结论 HAM是角朊细胞在体外培养的良好载体，对角朊细胞有明显的促增殖作用。

Morphological study on human amniotic membrane loading porcine keratinocytes for reconstructing epidermis

Objective To observe the morphological characteristics of human amniotic membrane(HAM)loading keratinocytes for reconstructing epidermis equivalent. Methods The primary cultures of Keratinocytes of Guizhou piglet, were primary cultured subcultured, expanded and bred on the stroma surface of HAM. The growth and proliferation of keratinocytes were observed under invert microscope each day. From the 3rd day to 8th day after culture, the growth of keratinocytes on HAM were examined under light microscope and electron microscope. Results Keratinocytes evidently adhered to the stroma surface of HAM after cultured for about 30 min. Most of keratinocytes were adhered to the stroma surface of HAM within 24 h. Keratinocytes formed monolayer and covered HAM completely within 3 d. Conclusion HAM is a good carrier of keratinocytes cultured on it in vitro , and able to promote the proliferation of keratinocytes significantly.
