摘    要:目的观察醉酒对大鼠重型颅脑创伤后脑组织水通道蛋白-4(AQP4)表达的影响。方法雄性Wistar大鼠70只,随机分为假手术(SO组)、颅脑创伤(TBI组)、乙醇预处理(ETH组)。ETH组给予白酒3g/kg灌胃。除SO组外,其余两组采用自由落体打击法制作颅脑创伤模型。分别于伤后不同时间点,采用干湿重法检测脑组织含水量,应用免疫组织化学法检测AQP4表达。结果与TBI组相比,ETH组脑组织含水量及AQP4表达水平显著升高(P〈0.05),且通过相关性分析发现,大鼠脑损伤后脑组织AQP4表达水平与脑水肿呈正相关(r=0.684,P=0.001)。结论醉酒加重颅脑创伤后脑水肿与其升高AQP4表达有关。

Abstract:Objective To observe the effects of drunkenness on Aquaporin-4 (AQP4)in rats after severe traumatic brain injury. Methods Seventy adult male Wistar rats were randomly divided into sham group (SO group),severe traumatic brain injury group (TBI group)and ethanol pretreatment group (ETH group). Subjects in ETH group were given ethanol 3 g/kg by intragastric administration. The experimental model was established by Feeney's method in the other groups except for SO group. Samples were harvested at different time points after injury to measure the water content of injured brain by dry-wet weight method and to inspect the expression of AQP4 by immunochemical method. Results Compared to TBI group,the water content and expression of AQP4 increased significantly in the ETH group (P 0.05). According to the correlation analysis,the expression of AQP4 was positively correlated with the water content (r=0.684,P=0.001). Conclusions Ethanol pretreatment may augment brain edema after severe traumatic brain injury by up-regulation of the expression of AQP4.
