5-羟色胺与体液免疫应答的相互作用

摘    要:应用定量溶血分光光度测定法(QHS)检测5-羟色胺(5-HT)对大鼠体液免疫应答的影响,并应用高效液相色谱-电化学检测法(HPLC-ECD)测定大鼠在体液免疫应答的高峰期,中枢和外周5-HT代谢的变化。结果表明:给大鼠腹腔注射5-HT的前体5-羟色氨酸(5-HTP)后,动物对抗绵羊红细胞(SRBC)的体液免疫应答减弱;腹腔注射5-HT的合成抑制剂对氯苯丙氨酸(P-CPA)后,免疫应答增强,在体液免疫应答的高峰日(免疫后第5天),下丘脑和脑干内5-HT的含量明显增加,5-HT的代谢产物5-羟吲哚乙酸(5-HIAA)的含量无显著变化;脾脏中5-HT的水平无明显升高,而5-HIAA含量显著增多;海马和胸腺内5-HT代谢没有明显改变。上述结果提示,5-HT与体液免疫应答之间存在着相互作用,即5-HT可能抑制免疫应答,而免疫应答又可加强中枢神经系统和外周胸腺、脾脏内5-HT的代谢。

Abstract:In this study,we determined the effect of 5-hydroxytryptamine(5-HT) on the humoral immune response of rat by the aid of the quantitative hemolysis spoectrophotometry(QHS) and observed the changes of central and peripheral 5-HT metabolism on peak day of the humoral immune response of rat with the help of the high-performance liquid chromatography with electrochemical detection(HPLC-ECD).The results are as follows: Systemic administration of the precursor of serotonin, 5-hydroxytryptoph an(5-UTP),resulted in the suppression of rats to sheep red blood cells(SRBC) response, and injection of para-chlorophenylalanine(p-CPA), which inhibits the biosynthesis of serotonin, markedly enhanced immune response. On day 5 after immunization, the content of 5-HT in the hypothalamus and brainstem significantly increased,and the level of 5-hydroxyindole acetic acid(5-HTAA) which is the metabolite of serotonin was markedly high in the spleen; the 5-HT metabolism in the hippocampus and thymus did not significantly changes. The results suggest that there exists an interaction between the serotonin and humoral immune response, that is,5-HT may inhibit the humoral immune response and the immune response may enhance the central and peripheral 5-HT metabolism.
