舒拉明对培养的人视网膜色素上皮细胞移行的影响

目的 观察舒拉明（suramin)对培养的人视网膜色素上皮细胞（retinal pigment epithelium,RPE)移行的影响。方法 在体外培养的RPE细胞的损伤模型中，加入不同浓度的舒拉明（1.5,15,150mg/L)观察12h,24h和48h3个时间点RPE细胞移行率。结果 无血清培养液中的RPE细胞有较弱的移行能力，含有100ml/L新生小牛血清的培养液可以显著刺激RPE细胞移行（P＜0．01），3种浓度舒拉明可以显著抑制100ml/L血清条件下的RPE细胞的移行，12h移行率分别对照组的82％、74％和46％、24h为92％，83％和52％、48h为92％，84％和55％这种作用呈剂量依赖性。结论 血清中的某些成分可以刺激RPE细胞的移行，舒拉明可以抑制血清的这种作用，为临床预防和治疗增殖性玻璃体视网膜病变（proliferative vitreoretinopathy,PVR)提供了新的用药思路。

Effects of suramin on migration of cultured human retinal pigment epithelium cells in vitro

Objective To investigate effec ts of suramin on migration of cultrued human retinal pigment epithelium(RPE)cells in vitro.Methods The cultured human RPE cells added with suramin(1 5,15, 150 mg/L)were measured in a in vitro wound healing model.The cell migration ra te was observed at hours 12,24 and 48 respectively.Results The serum stimul ated RPE migraton effectively(P<0 01).The serum induced migration was signifi cantly inhibited by suramin in a dose dependent manner at hours 12, 24 and 48. Compared with control groups,the migration rate was 82%,74% and 46% at hour 12,w as 92%,83% and 52% at hour 24,and was 92%,84% and 55% at hour 48.Conclusion Th e serum induced migration of RPE inhibited by suramin may be useful in devising effective treatment of proliferative virtreoretinopathy(PVR).
