阿霉素对不同C-erbB-2表达水平肺腺癌细胞株生长的影响

摘    要:目的:研究阿霉素对体外培养不同C-erbB-2表达水平肺腺癌细胞增殖能力和细胞周期的影响。方法:采用集落形成实验检测阿霉素对人肺腺癌细胞体外增殖能力的影响,采用SABC法检测阿霉素对体外培养人肺腺癌细胞系C-erbB-2表达水平的影响,采用流式细胞术检测阿霉素对体外培养肺腺癌细胞生长周期的影响。结果:阿霉素可显著抑制3株人肺腺癌细胞的生长,并可使SPC-A-1和LTEP-a-2肺腺癌细胞系C-erbB-2的表达下降,在SPC-A-1和LTEP-a-2肺腺癌细胞系,G2-M期百分率明显升高,而在A549肺腺癌细胞系则可见明显的凋亡峰。结论:阿霉素抑制肺腺癌细胞株SPC-A-1和LTEP-a-2的生长机制可能与降低C-erbB-2表达引起肺腺癌细胞G2阻滞有关,其抑制肺腺癌细胞株A549生长则可能与诱导细胞凋亡有关。

Abstract:Objective:To investigate the effect of doxorubicin on multiplication capacity of human lung adenocarcinoma cell lines with the expression of C-erbB-2. Methods:The expression of C-erbB-2 in human lung adenocarcinoma cell lines was tested by using SABC immunohistochemistry method. The mitogenic capacity was detected by colony-forming assay. The cell cycle status was determined by flow cytometry. Results:There were expressions of C-erbB-2 of different level in the three cell lines. The restrictive action of doxorubicin on the three cell lines was examined.Compared with the control the positive rate of p185 was markedly decreased in SPC-A-1 cell line and in LTEP-a-2 cell line treated with doxorubicin. The change in A549 cell line was of no significance.The percentages of G2/M phase in SPC-A-1 and LTEP-a-2 cell lines treated with doxorubicin were significantly increased. In contrast, the percentage of S phase in A549 was increased. Conclusion:1. In human lung adenocarcinoma cell lines, the expressing of C-erbB-2 oncogene was observed. 2.The anticancer effect of doxorubicin in SPC-A-1 and LTEP-a-2 human lung adenocarcinoma cell lines may correlate with decreasing the expressing of C-erbB-2 and arresting G2, and may induce appotosis in A549 cell lines.
