全反式维甲酸诱导分化的HL-60细胞表面凝集素受体的变化

摘    要:目的:了解人类早幼粒细胞性白血病细胞株(HL-60)细胞经全反式维甲酸诱导分化后各时相细胞表面5种凝集素受体的变化.方法:以刀豆凝集素(ConA)、麦胚凝集素(WGA)、花生凝集素(PNA)、蓖麻凝集素(RCA)、荆豆凝集素(UEA-1)共5种Na^125I-凝集素,标记经全反式维甲酸诱导分化的HL-60细胞,检测各时相细胞表面凝集素受体的变化.结果:HL-60细胞从幼稚分化发育至成熟各阶段中,细胞表面与UEA-1、WGA、ConA结合的凝集素受体有增多的趋势,诱导分化前后各凝集素放射活性比较有显著性差异(P<0.05),其中WGA放射活性在分化前后明显增加,而与PNA结合的凝集素受体在HL-60细胞分化过程中呈减少的趋势,在诱导分化后期减少最为明显,诱导分化前后放射活性比较有显著性差异(P<0.05).结论:D-半乳糖-N-乙酰氨基半乳糖结构主要表达于HL-60细胞早幼粒阶段,随着细胞的分化成熟而显著减少,其可能是肿瘤特异性抗原.HL-60细胞在经全反式维甲酸诱导分化过程中,细胞膜上含L-岩藻糖糖链结构增多,N-乙酰氨基葡萄糖结构增多和/或唾液酸化作用增强,并出现D-葡萄糖和/或D-甘露糖糖链结构.

Abstract:Objective: To investigate changes in the carbohydrate structures of HL-60 cell surface during AT- RA induced differentiation. Methods: Five kinds of Na125I - binding lectins, i. e. ConA, WGA, UEA- 1, PNA, RCA, were used to detect changes in the carbohydrate structure at various stages of differentiation. Results: The levels of ConA, UEA- 1, WGA receptors apparently increased, while the binding of PNA obviously decreased during HL - 60 cell differentiation. Conclusions: The D - Gal - N - GalNAc structures were mainly expressed in the early differentiation stages of HL- 60, and the amount of expression decreased significantly, when cell differentiation came to its mature stage. As a result, D - Glc or D - Man - associated carbohydrate structures on the cell surface could be seen quite obviously
