丹参酮ⅡA对兔胆管成纤维细胞的影响

摘    要:目的:观察丹参有效单体成分丹参酮ⅡA对兔胆管成纤维细胞增殖活性及胶原代谢的影响。方法:分离培养日本大耳白兔胆管成纤维细胞。将成纤维细胞随机分为对照组和丹参酮ⅡA组,用MTT法检测不同浓度丹参酮IIA溶液作用24、48、72h对其增殖活力的影响;应用Elisa法检测丹参酮ⅡA作用48h后其I型胶原分泌量的变化,采用RT-PCR方法检测丹参酮IIA对其基质金属蛋白酶(MMP)-9mRNA表达的影响。结果:与对照组相比,0.1、0.5、2.5μg/mL浓度的丹参酮IIA对兔胆管成纤维细胞增殖无明显影响(P0.05),10μg/mL浓度的丹参酮IIA作用72h可抑制胆管成纤维细胞增殖(P0.05),25μg/mL浓度的丹参酮IIA,24h即表现出对兔胆管成纤维细胞增殖明显的抑制作用(P0.05),且随着药物作用时间的延长,抑制作用增强。丹参酮ⅡA组成纤维细胞Ⅰ型胶原分泌水平较对照组显著降低(P0.01)。其MMP-9mRNA的表达较对照组明显升高(P0.05)。结论:丹参酮ⅡA能够抑制良性狭窄胆管成纤维细胞增殖及胶原的分泌,能诱导MMP-9mRNA的表达而促进胶原降解,从而抑制胆道瘢痕的形成。

The Influence of Tanshinone IIA on Rabbit Bileduct Fibroblasts

Abstract:Objective This study is aimed to examine the effects of Effective components of Salvia- Tanshinone ⅡA on the fibroblasts of rabbit benign stricture bile duct proliferation collagen metabolism. Methods Fibroblasts cells were separated and cultured from Japan white rabbits bile duct. They were divided into control solution and tanshinone ⅡA group. Detection the influence of different concentrations tanshinone ⅡA on FB proliferation at 24h, 48h, 72h by MTT and the levels of collagen type I quantitatively by ELISA method; Determination of the mRNA expression of MMP-9 by RT-PCR method. Results Compared with control group, when the concentration of tanshinone IIA is 0.1 μg/mL、0.5 μg/mL、2.5 μg/mL, it's had no significant effect on rabbit bile duct fibroblasts(P 0.05); when the concentration of tanshinone IIA is 10 μg/mL, 72 h later show that inhibit fibroblasts proliferation(P 0.05), when the concentration of tanshinone IIA is 25 μg/mL, 24 h show that inhibit fibroblasts proliferation(P 0.05), and as the extension of time and the increase in drug concentration, the role of inhibition enhanced; The levels of fibroblast secret collagen type Ⅰ of Tanshinone IIA group is lower than the control group. The expression of MMP-9mRNA of Tanshinone IIA group is significantly higher than the control group. Conclusion Tanshinone IIA effectively inhibit benign stricture of bile duct fibroblasts proliferation and collagen type Ⅰ secretion, and tanshinone IIA can induce the expression of MMP-9mRNA, promote collagen degradation, thereby inhibiting the formation of bile duct scar.
