人参皂苷Rg1/ADSCs辅以透明质酸为基质对兔颞下颌关节骨关节病的改善作用

摘    要:目的:探讨人参皂苷Rg1/ADSCs辅以透明质酸为基质对兔颞下颌关节骨关节病的治疗效果。方法:32只新西兰大白兔随机分为空白组、模型组、对照组、实验组,每组8只。通过关节腔内注射木瓜蛋白酶法建立骨关节病模型,对照组与实验组造模成功2周后开始药物治疗。对照组关节上腔注射人参皂苷Rg1/ADSCs悬液0.6mL,实验组注射透明质酸/人参皂苷Rg1/ADSCs复合物0.6mL,每周1次。各组动物分别于用药第3、9周后处死,每次16只,每组4只。制备标本,行髁突软骨扫描电镜、组织学观察及实时定量PCR检测,采用SPSS17.0软件包对数据进行方差分析及t检验。结果:扫描电镜与组织学观察可见,实验组软骨病变区改善情况优于对照组。与模型组比较,实时定量PCR结果显示,实验组MMP-13表达与空白组相对持平但低于对照组,差异有显著性(P<0.05);实验组TIMP-1表达与空白组相对持平但高于对照组,差异有显著性(P<0.05)。结论:人参皂苷Rg1/ADSCs辅以透明质酸为基质对改善兔颞下颌关节骨关节病有明显作用。

Abstract:PURPOSE: To explore the therapeutic effect of ginsenoside Rg1/ ADSCs supplemented with hyaluronic acid as matrix on rabbits with temporomandibular joint osteoarthritis(TMJ-OA).METHODS: Thirty-two New Zealand white rabbits were randomly divided into four groups: blank group,model group,control group and experimental group,8 in each group.Osteoarthritis model was established by intra-articular injection of papain.Two weeks after successful model building,medication was given for the rabbits in control group and experimental group.0.6 mL ginsenoside Rg1/ADSCs suspension was injected into the superior joint space for rabbits in the control group,once a week;0.6 mL ginsenoside Rg1/ADSCs hyaluronic acid complex was injected for rabbits in the experimental group,once a week.Animals in each group were sacrificed 3 and 9 weeks after drug administration,respectively(16 for each time;n=4).Specimens were prepared for scanning electron microscopy(SEM),histological observation and real-time quantitative PCR(qPCR) of the condylar cartilage.The data was analyzed by analysis of variance and Student’s t test using SPSS 17.0 software package.RESULTS: Improvements in the area of cartilage lesions in the experimental group were better than in the control group by SEM and histological observation.Compared with the model group,the results from qPCR showed that the expression of MMP-13 in the experimental group was similar to the blank group,but was lower than the control group.The difference was statistically significant(P〈0.05).The expression of TIMP-1 in the experimental group was similar to the blank group,but was higher than the control group.The difference was statistically significant(P〈0.05).CONCLUSION: Ginsenoside Rg1/ADSCs supplemented with hyaluronic acid as matrix plays a significant role in improving TMJ-OA in rabbits.
