摘    要:目的 研究经脂多糖处理后人胚肺成纤维细胞中糖皮质激素受体的表达及地塞米松对其的影响.方法 人胚肺成纤维细胞经脂多糖处理后,给予地塞米松进行干预,行细胞病理学检查及细胞增殖实验,检测培养细胞上清液中羟脯胺酸含量,采用免疫组织化学及Western blot法检测细胞中糖皮质激素受体的表达.结果 应用地塞米松后,人胚肺成纤维细胞病理变化减轻,细胞的增殖明显减少,细胞上清液中羟脯胺酸含量显著下降,糖皮质激素受体表达增加,与脂多糖组比较差异均有统计学意义.结论 地塞米松可减轻脂多糖所致人胚肺成纤维细胞的纤维化反应,其作用机制可能与上调糖皮质激素受体有关.

Abstract:Objective To investigate the effects of dexamethasone on glueocorticoide receptor(GR) in human fetal lung fibroblast. Methods After activation of lipopolysaccharide (LP$), human fetal lung fibroblast was treated with dexamethasone. Histological examination was performed with HE staining. The level of hydroxyproline in supernanant was measured by digestion method. GR expression was detected by immunohistochemistry, Western blotting. Results Hydroxyproline content in supernatant decreased significantly in dexamethasone group in comparison with acute lung injury(ALI) group [(10.83±0.74) μg/ml vs (12.71±1.97) μg/ml] (P<0.01). GR level was significantly increased in dexamethasone group in comparison with ALI group. Conclusions Dexamethasone may be useful in inhibiting LPS-induced fibrosis in human fetal lung fibroblast. Dexamethasone may reduce fibrosis by increasing GR level in human fetal lung fibroblast.
