人轮状病毒体外持续感染机理的研究

摘    要:本文从人轮状病毒的产生、感染及增殖周期三个水平研究胰蛋白酶对人轮状病毒持续感染株的影响,以便探讨人轮状病毒体外持续感染的机理.研究结果发现,Mads-105细胞经胰蛋白酶处理后不能明显提高病毒产量,且人轮状病毒持续感染株经胰蛋白酶处理后和维持液中含胰蛋白酶对病毒在MA-104细胞中增殖亦无显著影响(P>0.05),从而表明人轮状病毒持续感柒株已失去对胰蛋白酶的依赖性.其对胰蛋白酶依赖性的突变可能是造成人轮状病毒体外持续感染的机理之一.

Abstract:In this paper, the effects of trypsin in production, infection and repli-cation of HRV-PI virus. the human rotavirus persistent infection strain, have been described. From the preliminary results, we found the yields of HRV-PI not significantly increased by the trypsin-pretreated Mads-105 cell line(P>0.05), the multiplication titers of trypsin-pretreated HRV-PI virus in MA-104 cell culture were not significantly different from that of the con-trol(P>0.05), and no effect of the trypsin content in the maintenance medium on the replication of HRV-PI in MA-104 cell was detected. These results indicate that HRV-PI iS a trypsin independent muttant which might be among the mechanisins of human rotavirus persistent infection in vitro.
