摘    要:目的 观察慢性肾功能不全 (CRF)患者树突状细胞 (DC)形态和功能的变化。方法 从 31例CRF患者和 2 5例健康人外周血中分离和培养DC ,观察DC的形态 ,用流式细胞仪检测DC表面标记CD1a ,CD4 0 ,CD80和CD83的表达 ,用MTT摄入法测定DC诱导混合性淋巴细胞反应 (MLR)的能力。结果 正常DC较CRF患者在形态上更为典型 ,不规则 ;CD1a ,CD4 0 ,CD80和CD83分子的表达水平较高 (P <0 0 5 ) ,诱导MLR的能力较强 (P <0 0 5 )。结论 CRF患者外周血DC处于不完全成熟状态 ,其免疫刺激能力较低。

Abstract:Objective To compare the differences of dendritic cell(DC) derived from healthy controls and patients with chronic renal failure(CRF) and to clarify the pathogenesis of immunodeficiency in chronic renal failure. Methods DC was isolated by density gradient centrifugation from peripheral blood monocytes harvested from 31 patients with different level of CRF and 25 healthy volunteers and was cultured in vitro. Their phenotypes, surface markers and stimulatory capatcities in mixed lymphocyte reactions were compared by microsopic observations, flow cytometry and mixed lymphocytes reaction combined with MTT incorporation. Results DC in CRF had a typical phenotype, lower surface expressions of CD40,CD80 and CD83(P<0.05), lower capacities to stimulate T cells in MLR(P<0.05). Conclusion These results reveal a specific defects of DC from CRF in their maturation and function, and may be related to the pathogenesis of immunodeficiency in CRF, suggesting that the induction of DC maturation and stimulatory capacity might be an approach to DC-based immunotherapy on CRF.
