| 摘 要： | 目的:探讨mag-1基因与肿瘤细胞转移的关系.方法:检测mag-1在人肺巨细胞癌高低转移细胞株PLA801D与PLA801C、肺癌高低转移细胞株A549与H1299及乳腺癌高低转移细胞株MCF-7与MDA-MB-231中mag-1的表达水平;检测构建的mag-1正反义真核表达载体转染细胞后mag-1表达水平的变化;将mag-1正义表达载体分别转染低转移细胞株PLA801C及MCF-7并筛选出稳定克隆;研究mag-1过表达对细胞形态、增殖、迁移、黏附及侵袭等生物学行为的影响.结果:高转移细胞株PLA801D中mag-1的mRNA及蛋白表达水平均显著高于低转移细胞株PLA801C中mag-1的表达水平,乳腺癌高低转移细胞株中mag-1的蛋白表达水平也有明显的差异;过表达mag-1细胞的形态及增殖能力没有发生改变,而细胞的黏附、迁移及侵袭能力增强.结论:mag-1可能促进肿瘤细胞的转移.
