电针预处理对局灶性脑缺血再灌注大鼠pSTAT3蛋白表达的影响

摘    要:目的:探讨电针预处理对大鼠脑缺血再灌注损伤后脑组织磷酸化STAT3( pSTAT3)蛋白表达的影响.方法:大鼠随机分为假手术组、模型组和电针预处理组,其中电针预处理组在造模前7天给予治疗,1次/天.局灶性脑缺血再灌注损伤大鼠模型的制备应用大脑中动脉线栓法,Western blotting检测缺血侧脑组织pSTAT3蛋白表达情况,TTC染色检测脑梗死体积.结果:缺血再灌注损伤24 h,电针预处理治疗组能明显缩小脑梗死体积,减少脑缺血大鼠pSTAT3蛋白的表达,与模型组比较均有显著性差异(P<0.05).结论:电针预处理能下调大鼠脑缺血再灌注损伤后pSTAT3的表达水平,明显缩小脑梗死体积,具有良好的神经保护作用.

Abstract:Objective:To investigate the effect of electroacupuncture pre - stimulation to pSTAT3 protein ex- pression in rats with the focal cerebral ischemia - reperfusion. Methods: The rats were randomly divided into fake operation group, model group and electro acupuncture pre - stimulation group. And the electro acupunc- ture pre - stimulation group was treated seven days before modeling, a day once. The model of focal cerebral is- chemia- reperfusioninjury was established by middle cerebral artery occlusion using modified filament. TFC stains was employed to evaluate cerebellar infarction area, and western blotting analysis was used to detect the expression of pSTAT3 protein. Results:After 24 hours'cerebral ischemia - reperfusion, the electro acupuncture pre - stimulation group is able to shorten cerebellar infarction area, decrease pSTAT3 expression of cerebral is- chemia rats and has significant difference than model group ( P 〈 0.05 ) . Conclusion: The electro acupuncture pre - stimulation is able to down regulate pSTAT3 protein expression of rats with the focal cerebral ischemia - reperfusion and significantly reduces the cerebellar infarction area, and has good function of nerve protection.
