摘    要:目的]研究P53通路相关蛋白在云锡矿粉诱导的细胞恶性转化过程中的变化情况。方法]采用免疫荧光细胞化学染色技术分别检测正常人支气管上皮细胞株(BEAS-2B)、矿粉作用后第25代细胞(BEAS-MD-25)及软琼脂克隆形成细胞(BEAS-MD-C)中P53、MDM2、P21、P16的表达。结果]P53、MDM2蛋白在BEAS-2B细胞中表达呈阴性,而在BEAS-MD-25及BEAS-MD-C细胞中均呈阳性;P21、P16蛋白在BEAS-2B细胞中表达呈阳性,随细胞转化进程表达不断下降;在BEAS-MD-25细胞中呈弱阳性;在BEAS-MD-C中呈阴性。结论]转化细胞中P53、MDM2、P21、P16的表达异常,提示其P53通路已发生改变,第25代细胞P53蛋白的阳性表达,提示P53基因异常可能是细胞发生恶性转化的关键步骤之一。

Abstract:Objective] To investigate the expression of P53 pathway proteins in malignant transformation cells of human bronchial epithelial cells induced by Yunnan tin mine dust.Methods] Human bronchial epithelial cells(BEAS-2B),the 25~(th) generation cells treated with mine dust(BEAS-MD-25)and selected in soft agar medium clonecells(BEAS-MD-C)were examined for P53,MDM2,P21 and P16 proteins expression by Immunofluorescence eytochemistry staining techniques.Results] The expressions of P53 and MDM2 were negative in BEAS-2B and were positive in BEAS-MD-25 and BEAS-MD-C cells.The expressions of P21 and P16 were positive in BEAS-2B.Along with the cells transformation the expressions of P21 and P16 were reduced,which were weak-positive in BEAS-MD-25 and negative in BEAS-MD-C.Conclusion]The abnormal expression of P53,MDM2,P21 and P16 proteins in transformation cells indicates the P53 pathway may be changed.The positive expression of P53 protein in the 25~(th) generation cells indicates the abnormality of P53 may be one of the key steps in cell malignant transformation.
