缬沙坦对人脐动脉平滑肌细胞妊娠相关蛋白-A、基质金属蛋白酶-9表达的影响

目的探讨缬沙坦对人脐动脉平滑肌细胞PAPP-A、MMP-9表达的影响。方法分别在10^-5mol／L的Ox—LDL的基础上，给予AngⅡ以及AngⅡ、缬沙坦混合培养人脐动脉平滑肌细胞，以普通培养基培养为对照，应用RT—PCR法检测PAPP—A、MMP-9的表达量。结果AngⅡ组PAPP-A与MMP-9的表达量均显著高于对照组、Ox—LDL及缬沙坦组（P〈0．01），缬沙坦组PAPP—A的表达量高于对照组（P〈0．01），而与Ox—LDL组无明显差异（P〉0．05）．缬沙坦组MMP-9的表达量与Ox—LDL组及对照组均无明显差异（P〉0．05）。结论AngⅡ可促进人脐动脉平滑肌细胞PAPP—A和MMP-9的表达，缬沙坦可以抑制AngⅡ的这种作用。

The Influence of Valsartan on PAPP-A, MMP-9 Gene Expression in Human Umbilical Artery Smooth Muscle Cell

Objective To study on the effect of Valsartan on the expression of the PAPP-A and MMP-9 in human umbilical arterial smooth muscle cell. Method Under the condition of Ox-LDL, HUVSMC was cultured with Ang Ⅱ , or Ang Ⅱ plus Valsartan. Cultures with no Ox-LDL were used a control. The expressions of PAPP-A and MMP-9 were determined using RT-PCR and ELISA. Result The expression levels of PAPP-A and MMP-9 in the culture with Ang Ⅱare higher than those in the other groups. The expression level of PAPP-A of Valsartan group is higher than that of the control group, but no difference from that in the Ox-LDL group. The expression level of MMP- 9 of the Valsartan group has no significant difference compared with that in the Ox-LDL group and the control group. Conclusion Ang Ⅱ induces the secretion of PAPP-A, MMP-9, while Valsartan can counter act the Ang Ⅱaction.
