慢性HCV感染者血清白细胞介素-10水平的变化及其意义

目的探讨白细胞介素(IL)-10在丙型肝炎病毒(HCV)致病过程中的作用。方法用酶联免疫吸附法(ELISA)检测44例慢性HCV感染者、30例健康对照者血清中IL-10的水平。用定量PCR法检测血清HCV-RNA水平。结果慢性HCV感染者的血清IL-10水平明显高于健康人(P<0.001)并且与丙氨酸氨基转移酶(ALT)的水平呈正的直线关系(P<0.01);慢性HCV感染者中,肝硬化组IL-10的水平较慢性丙型肝炎组有所降低(P<0.01);慢性HCV感染者的IL-10水平与血清HCV-RNA水平无相关性(P>0.05)。结论机体的IL-10应答参与了慢性HCV感染者的肝脏损害过程,并且可能是影响丙型肝炎慢性化的因素之一;IL-10产生的降低可能与慢性HCV感染者的肝纤维化和肝硬化有关。

Determination of serum IL-10 level in patients with chronic HCV infection and its significance in pathogenesis of chronic HCV infection

To investigate the role of interleukin (IL)-10 in the pathogenesis of chronic hepatitis C virus (HCV) infection. Serum levels of IL-10 in 44 patients with chronic HCV infection and 30 healthy individuals were determined by enzyme-linked immunoabsorbent assay (ELISA). Serum HCV-RNA levels were determined by quantitative polymerase chain reaction (PCR). Serum IL-10 level was significantly increased in patients with chronic HCV infection as compared with that in healthy individuals (P <0.001). The IL-10 levels in patients with chronic HCV infection correlated with their ALT levels (P <0.01). The IL-10 level did not correlate with HCV viral loads. IL-10 level was significantly decreased in patients with HCV associated cirrhosis as compared with that in patients with chronic hepatitis C (P <0.01). Conclusion] Host IL-10 response may be involved in the pathogenesis of chronic HCV infection and the defect production of IL-10 may be responsible for the progression to cirrhosis in patients with chronic HCV infection.
