关节腔注射青藤碱对兔膝骨性关节炎血清NO等自由基代谢的影响

摘    要:目的 观察关节腔内注射青藤碱(SIN)对兔膝骨性关节炎(OA)血清中自由基代谢的影响,以探讨其防治OA的作用机理.方法 将38只兔随机分为正常组(A)、模型组(B)、透明质酸钠组(C)和青藤碱组(D).A组不予任何处理,B,C和D三组通过关节腔注射木瓜酶法诱导OA模型后,分别用生理盐水、透明质酸钠和青藤碱干预,5周后检测并比较各组血清中一氧化氮(NO)、超氧化物歧化酶(SOD)和丙二醛(MDA)的含量.结果 B组血清NO和MDA含量分别为75.27±2.08 μmol/L和3.54±0.17 nmol/ml,均明显高于A组62.64±3.33 μmol/L和2.88±0.14 nmol/ml,差异有统计学意义(P值均<0.01);而B组SOD含量205.74±22.08 U/ml明显低于A组267.59±17.09 U/ml,差异有统计学意义(P<0.01).D组血清NO和MDA含量分别为69.29±1.51 μmol/L和3.09±0.16 nmol/ml,均明显低于B组,差异有统计学意义(P值均<0.01),低于C组72.69±1.84 μmol/L和3.33±0.22 nmol/ml,差异有统计学意义(P值均<0.05);而SOD含量245.46±11.25 U/ml,明显高于B组,差异有统计学意义(P<0.01),高于C组224.03±9.90 U/ml,差异有统计学意义(P<0.05).结论 关节腔注射青藤碱可以降低血清中NO,MDA含量,提高SOD活性,从而保护关节软骨,对OA起到防治作用.

Effect of Inject Sinomenine to Articular on the Metabolism of NO and Other Free Radicals in the Serum of Rabbits with Genicular Osteoarthritis

Abstract:Objective To observe the effect of intra-artieular injection of sinomenine(SIN) on the metabolism of free radicalsin the serum of rabbits with genicular osteoarthritis(OA),in order to investigate the mechanism of its prevention and treat-ment of OA. Methods The 38 rabbits were randomly divided into 4 groups:normal group(A) ,model group(B), sodium hya-luronate group(C) and sinomenine group(D). B,C, D group were established genicular osteoarthritis model by intra-articularinjectionof papain enzyme and were treated with saline,sodium hyaluronate and sinomenine respectively except for A group.5 weeks after treatment,investigate the level of the NO, SOD and MDA in the serum of the rabbits. Results The level ofNO (75.27±2.08) μmol/L and MDA (3.54±0.17) nmol/ml in B group were both higher than that of the A group (62.64±3.33) μmol/L and (2.88±0. 14) nmol/ml,there were distinct difference(both P〈0.01) ,while the level of SOD (205.74±22.08) U/ml was significantly lower than that of the A group (267.59± 17.09) U/ml (P〈0.01). The level of NO(69.29±1.51) μmol/L and MDA(3. 09±0.16) nmol/ml in D group were both significantly lower than that of the B group(both P〈0.01) ,distinct lower than C group (72.69±1.84) tzmol/L and (3.33±0.22) nmol/ml (both P〈0.05) ,whilethe level of SOD (245.46±11.25) U/ml was significantly higher than that of the B group (P〈0.01) ,and distinct higherthan C group (224.03±9.90) U/ml (P〈0.05). Conclusion Articular injection of sinomenine can decreased the levels ofNO and MDA and increase the activity of SOD in the serum. So it can protect the articular cartilage and play a preventiveeffect of genicular osteoarthritis.
