鼻渊舒口服液抑制鼻黏膜成纤维细胞生长的实验研究

目的 本研究旨在观察中药鼻渊舒口服液对慢性鼻炎鼻黏膜中成纤维细胞的作用,探讨鼻渊舒口服液治疗慢性鼻炎的可能机理.方法 取慢性鼻炎患者的鼻黏膜,经传代培养后获得成纤维细胞.采用四氮唑盐(MTT)法对鼻渊舒口服液抑制成纤维细胞的生长进行研究,计算抑制率,绘制药物浓度?抑制率曲线.采用透射电子显微镜对细胞的超微结构进行观察.结果 鼻渊舒口服液显著抑制鼻黏膜成纤维细胞的生长,其抑制作用随药物的浓度和作用的时间的增加而增强.经过该药处理后,可以观察到鼻黏膜成纤维细胞在超微结构上发生凋亡改变.结论 中药鼻渊舒口服液可以导致鼻黏膜成纤维细胞发生凋亡,其对鼻黏膜成纤维细胞的抑制作用具有浓度依赖性和时间依赖性.

Experimental study on the effect of Biyuanshu oral liquid on the proliferating activity of nasal fibroblasts in vitro

Objective To observe the effect of Biyuanshu oral liquid(BOL) on the proliferating activity of fibroblasts in vitro in the nasal mucosa of patients with chronic rhinitis to investigate the possible mechanisms responsible for the therapeutic effect of BOL on this lesion.Methods Fibroblasts were collected from the nasal mucosa samples taken from the cases with chronic rhinits via culturing for several generations in vitro.Then,MTT(methyl-thiazolyl-tetrazoliun) method was utilized to measure the proliferating activity of these cells to plot concentration-inhibition curve via calculating the inhibitory rate of cells,and transmission electron microscopy was made to observe the morphological changes of these cells at ultrastructural level.Results It was shown that BOL could significantly inhibit the proliferating activity of nasal fibroblasts and it held the characters of strengthened inhibitory effect depending upon both parameters of the medicinal concentration and its affecting time length.The cells showed apoptotic changes at ultrastructural level following the treatment of BOL.Conclusions BOL can induce apoptosis to nasal fibroblasts and this kind of effect hold concentration-dependent and time-dependent characters for the medicine.
