Abstract:Objective: To study the effect and mechanism of different sodium on bone growth in immature rats. Methods: Thirty-six 5-weeks-old female SD rats were randomly divided into 3 groups: low dose sodium group (LDS, 0.13% NaCl), normal dose group (NDS, 1% NaCl), and high dose sodium group (HDS, 8% NaCl). All doses of sodium were given orally by food for 60 days. The left tibia (proximal tibia metaphyses, PTM), middle (middle part of tibia shaft, TX) were taken for bone histomorphometry in the experiment. Results: In PTM, the percent trabecular bone area (BV/TV) was increased in LDS (p<0.05) and not in HDS compared with NDS group. The trabecular thickness was decreased in HDS compared with LDS group. Dynamic data showed that the bone formation rate (BFR/TV) was higher, number of osteoclast and surface perimeter of osteoclast were lower in LDS group than NDS group. No significantly different were founded between HDS group and NDS group, and No significantly different were founded between HDS group and LDS group. In Tx, There were no statistically changes in cortical static parameter in three groups. But dynamic data showed that HDS enhanced endosteal bone resorption, while LDS depressed endosteal bone resorption compared with normal group. In contrast to LDS, HDS was showed to increase percent labeling parameter and BFR in endosteal surface. Conclusion: The different effect on bone group by different concentration of sodium. Low dose sodium is benefit to bone group in immature SD female rat by increasing bone formation and decreasing bone resorption; but the effect of HDS on immature rat is vary, which may be due to individual variation.
