摘    要:研究Wilms瘤基因在白血病发病中的作用机制。方法:采用反义、Northern blot和流式细胞技术分析了硫以义WT-1基因脱氧寡核苷酸和硫代反义c-myb基因对U937细胞增殖和基因表达的影响。结果:μmol/L WT1反义脱氧寡核苷酸处理的U937细胞WT1蛋白表达下降37%,c-myb mRNA水平上升不明显:10μmol/L c-myb mRNA水平上升明显,同时Myb蛋白水平上升46%

Abstract:Objective: To elucidate a role for the Wilms tumor gene (WT1) in leukemogenesis. Methods: We investigated the effect of phosphorothioate antisense oligodeoxynucleotides (AN-S-ODNs) specific to WT1 gene on the expression of c-myb gene in U937 cells. Northern blot and flow cytometry technologies were used to detect mRNA and protein levels, respectively. Results: 5umol/L AN-S-ODNs for WT1 mRNA significantly inhibited WT1 protein expression in U937 cells at 37% (p<0.01). 10 umol/L AN-S-ODNs inhibited WT1 protein expression in U937 cells at 87% (p<0.01). And increased c-myb expression both at mRNA and protein level (46%, p<0.01), but no effect on WT1 expression in U937. Conclusions: Wilms tumor gene regulates the expression of c-myb gene expression in U937 cells.
