多囊卵巢综合征大鼠子宫内膜胰岛素受体和胰岛素生长因子-1表达的研究

目的观察多囊卵巢综合征(PCOS)大鼠子宫内膜胰岛素受体(IR)和胰岛素生长因子(IGF-1)的表达以及应用二甲双胍后子宫内膜IR和IGF-1表达的改变。探讨影响PCOS大鼠子宫内膜容受性的原因。方法选取6周龄清洁级雌性Wistar大鼠55只,随机分为对照组15只,模型组40只。应用来曲唑灌胃建立PCOS大鼠模型,造模成功后,模型组40只随机抽取10只作造模效果评价,其余30只随机分为二甲双胍治疗组15只和PCOS组15只分别喂服二甲双胍和生理盐水。14 d后麻醉大鼠并取材,光镜观察子宫内膜的情况,免疫组织化学法测定3组子宫内膜IR和IGF-1的表达水平。结果 PCOS组大鼠子宫内膜腺上皮和间质中IR蛋白表达明显低于对照组(P<0.05),IGF-1的表达明显高于对照组(P<0.05)。治疗组IR表达较PCOS组显著增高(P<0.05)但仍低于对照组(P>0.05),治疗组IGF-1表达比PCOS组降低(P<0.05)与对照组相比仍然较高(P<0.05)。结论 PCOS大鼠子宫内膜IR和IGF-1均与对照组有明显的差异。通过二甲双胍治疗后可部分逆转此变化,说明IR和IGF-1与PCOS子宫内膜改变有相关性。

Expression of insulin receptor insulin growth factor-1 in endometrial of rats with polycystic ovary syndrome

Objective To observe the expression of insulin receptor（IR） and insulin-like growth factor-1（IGF-1） of endom etriumin rats with polycystic ovarian syndrome（PCOS）,and the change of IR IGF-1 of endom etrium in rats after therapy with metformin.investigate the mechanism affect in endometrial receptivity of rats with PCOS.Methods Set up an animalmodel by using the non-steroidal aromatase inhibitor-letrozole.40 female Wistar rats of sixweeks age were randomly divided into three groups： the control group（n=15）,PCOS model group（n=15）,metformin in treatment group（n=15）.control group feed normally the latter group followed by metformin and normal saline respectively.After 14 days treatment,the rats were anesthesiaed and sacrificed.Endometrial condition were observed by light microscope.Using the immunohistochemical staining technique detected the expressed level of IR and IGF-1.Results The expression of insulin receptor（IR） proteins of endometrial in PCOS rats were significantly lower（P0.05） than those in control group,the expression of（IGF-1） protein in PCOS rats was higher than that in control group（P0.05）.The expression of IR in the treatment group increased（P0.05）,but was still lower than that in control group（P0.05）.However,compared with PCOS group,the expression of IGF-1 protein in treatment group was decreased（P0.05）,but was stilL higher than that in control group（P0.05）.Conclusion IR and IGF-1 of endom etrium in PCOS group compared with control group were obviously differant through the metformin treatment may reversal the situation.Conclude that IR and IGF-1 with change of PCOS endometrial have correlation.
