摘    要:目的 研究尾加压素Ⅱ(UⅡ)对大鼠心室肌细胞L-型钙电流(Ica-L)的影响。方法 利用全细胞膜片钳技术,观察给予不同浓度的UⅡ灌流后,大鼠心室肌细胞Ica-L的变化。结果 运用全细胞膜片钳技术,在单个心室肌细胞上给予不同浓度的UⅡ(10-9~10-5mol/L)灌流5min后,Ica-L峰值下降分别为(6.7±1.8)pA/pF,(5.9±2.0)pA/pF,(5.4±2.2)pA/pF,(4.5±2.0)pA/pF,(3.8±1.8)pA/pF,与对照组(8.0±1.2)pA/pF相比,差异有统计学意义(P<0.05)。结论 UⅡ呈浓度依赖性地抑制大鼠心室肌细胞Ica-L强度;UⅡ可能通过抑制心室肌细胞Ica-L而发挥其心功能抑制作用。

Abstract:Objective To investigate the effects of Urotensin Ⅱ (UⅡ) on L-type calcium current (Ica-L) in rat ventricular myocytes. Methods Whole-cell patch clamp technique was employed to investigate the changes in rat cardiac myocyte Ica-L after perfusion using different concentrations of UⅡ. Results Whole-cell patch clamping showed that the peak values of Ica-L in single cardiac myocytes were reduced to (6.7±1.8) pA/pF, (5.9±2.0) pA/pF, (5.4±2.2) pA/pF, (4.5±2.0) pA/pF and (3.8±1.8) pA/pF, respectively, at five minutes after perfusion using different concentrations of UⅡ (10-9~10-5 mol/L), which were statistically different (all P0.05) as compared with control group (8.0±1.2) pA/pF]. Conclusion UⅡ inhibits Ica-L in rat cardiac myocytes in a dose-dependent manner, and thereby exhibits inhibitory effects on cardiac function.
