5－Fu佐剂包裹物的抗肿瘤作用的实验研究

本文作了福氏完全佐剂（Ｆｒｅｕｎｄ’ｓｃｏｍｐｌｅｔｅａｄｊｕｖａｎｔ,ＦＣＡ）包裹５－Ｆｕ的抗肿瘤作用的实验研究。在对小鼠Ｓ１８０肿瘤（腹水型）,ＥＡＣ肿瘤（腹水型）及Ｈ２２肝癌（腹水型）荷瘤鼠的治疗实验中,一次性腹腔注射２ｍｇ～６ｍｇ的５－Ｆｕ（相当于７０公斤人给予７７８～２３３４ｍｇ的剂量）均不能显著延长荷瘤鼠的存活期,盐水ＦＣＡ亦无治疗作用,而５－ＦｕＦＣＡ却显示出显著的治疗效果。４次实验,５－ＦＵ－ＦＣＡ治疗组荷瘤鼠平均存活期３０．６天～４８．３天,均显著长于同次实验的各对照组,且每次实验都有２／８至３／８的荷瘤鼠可完全治愈。

Experimental Studying on Antitumor Effects of 5-Fu Entrapped by FCA

FCA (Freund's Complete Adjuvant,FCA)is usually used to entrap antigen to immunize animal, We entrapped 5-Fu with FCA to slow their release in vivo and by means of this to reduce their side effects and to augment their antitumor effects. We called these FCA entrapped 5-Fu,5-Fu-FCA. The results showed that:the average survival time of 5180, EAC and H22 tumor bearing mice treated with 5-Fu was 18. 3±1. 6, 17. 9±1. 9 and 19. 1±3. 6 days respectively, none of them were cured, while that of S-Fu-FCA treated groups was 48. 3±21. 1,30. 6±2. 3 and 33. 4±21. 6 days and 3/8, 2/8, Z/8 of the tumor bearing mice cured respectively (vs S-Fu treated groups,Ｐ＜0. 0001). We conclude that:(1) the adverse effects of 5-Fu could be significantly reduced by means of FCAentrapping; (2)the antitumor effects of 5-Fu could be augmented by means of FCA entrapping.
