青光眼患者小梁网中一氧化氮合酶的表达

摘    要:目的 检测正常人和急性闭角型青光眼(acute angle-closure glaucoma,AACG)患者小梁网中诱导型一氧化氮合酶(inducible nitric oxide synthase,iNOS)和内皮型一氧化氮合酶(endothelia nitric oxide synthase,eNOS)的表达,探讨小梁网中的一氧化氮合酶的变化及与青光眼的关系.方法 取5例正常人及15例AACG患者小梁网组织标本,应用免疫组织化学方法检测小梁细胞eNOS和iNOS的表达,并采用计算机图像分析系统对检测结果进行分析.结果 正常人的小梁网组织标本有eNOS的表达,阳性细胞分布均匀,iNOS呈弱阳性或不表达.AACG患者小梁网组织标本中,eNOS和iNOS均呈阳性表达,iNOS主要分布在邻管组织和Schlemm管.iNOS阳性表达比正常对照组增强(P<0.05),而eNOS阳性表达比正常对照组减弱(P<0.01).结论 AACG患者小梁网细胞eNOS表达减少而iNOS表达增多且主要分布在邻管组织、Schlemm管,提示一氧化氮合酶(nitric oxide synthase,NOS)在青光眼发病中起重要作用,是导致或加重青光眼的因素之一.

Abstract:Objective To identify expression of inducible nitric oxide synthase(iNOS) and endothelia nitric oxide synthase(eNOS) expression in trabecular meshworks(TM) of normal control and patients with acute angle-closure glaucoma(AACG),and investigate the distribution of iNOS,eNOS in the TM and the relationship with AACG.Methods TM specimens were taken from 5 normal persons and 15 patients with AACG.The expression of iNOS and eNOS were studied with SP immunohistochemical technique.The results were analyzed with computer-image-analysis system.Results The trabecular meshwork cells in all the 5 normal persons expressed eNOS.The positive cells existed in the TM evenly,while the expression of iNOS are weak positive or absent.In TM of patients with AACG,the TM cells expressed both eNOS and iNOS,iNOS was expressed mainly in the adjacent trabecular meshwork and schlemm' channel.The stained intensity of iNOS was stronger than that of the normal control group(P<0.05).The positive expression of eNOS is more reduced obviously as compared with that of eyes with the normal control group(P<0.01).Conclusion In the TM of patients with AACG,the expression of eNOS are reduced while iNOS are higher than that of normal eyes.And iNOS is mainly expressed in the adjacent trabecular meshwork and Schlemm's channel.It suggests that NOS played important role on glaucoma and it may induce or aggravate glaucoma.
