生长抑素对白血病患者骨髓细胞CD34抗原表达的影响

摘    要:目的研究生长抑素八肽类似物奥曲肽(Octreotide,SMS;商品名善得定)对白血病患者骨髓单个核细胞CD34抗原表达的影响,探讨生长抑素对白血病细胞的调控作用.方法采用细胞培养、免疫细胞化学技术及图像分析系统,随机计数CD34阳性细胞数,观察分析SMS在白血病患者骨髓单个核细胞中CD34抗原表达的分布和含量.结果在相差倒置显微镜下,加入SMS组与对照组比较,细胞碎片增多,基质细胞减少;CD34抗原主要表达于细胞膜和核膜;SMS组阳性细胞数显著少于对照组(P<0.01);图像分析光密度值(OD值)SMS组明显低于对照组(P<0.01).结论在体外SMS可减少造血干/祖细胞特异抗原(CD34)的表达.提示SMS对白血病细胞的生长具有负性调控作用.

Abstract:Objective To study the effect of octapeptide somatostatin analogue (Octreotide,SMS) on the expression of CD 34 antigen in leukemic bone marrow mononuclear cells and to investigate the regulatory mechanism of somatostatin on leukemia cells.Methods The leukemic bone marrow mononuclear cells were treated with SMS in vitro.The CD 34 positive cells were counted and the expression of the distribution and concentration of CD 34 antigen in culture were observed with immunocytochemistry and imagine analytic system.Results In SMS group cellular fragments of leukemic bone marrow cells increased while stromal cells decreased under the inverted phase contrast microscope;the CD 34 antigen expression was mainly on cell membrane and nuclear membrane,and CD 34 positive cells were reduced significantly compared with the control group (P<0.01).The optical density (OD) reduced significantly in 10 -6 mol/L SMS group (P<0.01).Conclusion SMS reduces the expression of CD 34 antigen on hemopoietic stem cell and hemopoietic progenitor in vitro,which suggests that SMS plays an negtive role in cell growth of leukamia.
