不同剂量碱性成纤维细胞生长因子对神经侧枝再生的影响

摘    要:目的: 探讨碱性成纤维细胞生长因子 (basicFibroblastGrowthFactor, bFGF) 能否促进神经端侧缝合后的侧枝再生, 其作用是否存在量效关系。方法: 15只Wistar大鼠随机分为A、B、C3组, 切断一侧腓总神经并将其远端与外膜开窗的胫神经做端侧缝合。A、B2组于术后 1周内每天在腹腔内注射生理盐水稀释的bFGF, A组剂量 4 000u/kg, B组剂量 2 000u/kg, C组注射等量生理盐水做对照。术后 3个月取缝合口远端的腓神经做组织学检查。结果: 应用bFGF后再生神经纤维数目、有髓纤维截面积和髓鞘截面积显著大于生理盐水对照组 (P<0. 05 ); 而A组上述三项指标均显著大于B组 (P<0. 05 )。结论: 神经端侧缝合后早期应用外源性bFGF可促进未受损神经的侧枝发芽, 其作用与剂量呈正相关。

Abstract:Objective:To investigate the effects of basic fibroblast growth factor (bFGF) in collateral sprouting of intact nerve after end to side neurorraphy.Method:Fifteen Wistar adult rats were used in this study.The common peroneal nerves at one side were sectioned at the level of knee joint,and their distal ends were sutured laterally to the tibial nerves after removal of a 1 mm diameter window in the epineurium.Then the rats were divided into Group A,Group B and Group C randomly,bFGF were injected intraperitoneal to Groups A and B daily for 7days,Group A received 4000U kg l and Group B received 2000U kg 1 .The control Group C received normal saline.Three months postoperatively,the peroneal nerves distal to the neurorrhaphy site were harvested and underwent morphology analysis.Result:The number of regenerative nerve fibers,cross area of myelinated fibers and myelin in Groups A and B were significant larger than that in Group C.All parameters tested in Group A were significant larger than that in Group B.Conclusion: bFGF can promote the collateral sprouting of intact nerves after end to side neurorraphy,and the effect is correlated with the dosage positively.
