肿节风总黄酮对溴酸钾诱发小鼠肾损伤的保护作用

摘    要:目的 探讨肿节风总黄酮对溴酸钾诱发小鼠肾损伤的保护作用.方法 200mg· kg-1溴酸钾腹腔注射诱发小鼠肾损伤模型,观察肿节风总黄酮给药后对肾损伤的保护作用.采用HPLC法测定小鼠血清中肌酐含量及肾组织中谷胱甘肽含量,分光光度法测定小鼠肾组织中丙二醛含量和谷胱甘肽过氧化物酶活力,Griess反应法测定血浆及肾组织的一氧化氮含量.结果 与肾损伤模型组相比,肿节风总黄酮可以有效降低血清肌酐水平和肾组织中丙二醛含量,对血浆及肾组织中一氧化氮水平的升高也有一定的改善作用,同时可提高肾损伤小鼠肾组织的谷胱甘肽及谷胱甘肽过氧化物酶水平.结论 肿节风总黄酮对溴酸钾诱发的小鼠肾损伤具有一定的保护作用,其作用机制可能与肿节风总黄酮通过清除自由基缓解溴酸钾诱发的肾组织氧化应激状态有关.

Abstract:OBJECTIVE To investigate the protective effects of total flavonoids of Sarcandra glabra(Thunb.)Nakai(TFS) on potassium bromate(KBrO3)-induced kidney damage in mice.METHODS The kidney damage model was established by receiving a single ip administration of 200mg·kg-1 KBrO3 in mice.Serum creatinine level and the contents of glutathione(GSH) in kidneys were determined by high performance liquid chromatography(HPLC).Spectrophotometric method was performed to detect the levels of malondialdehyde(MDA) and glutathione peroxidase(GSH-PX) activity in kidneys.Griess method was performed to detect nitric oxide(NO) levels in plasma and kidneys.RESULTS Compared with the model group,TFS could effectively decrease serum creatinine level and the contents of MDA in kidneys.NO levels in plasma and kidneys decreased in the same schedule.In contrast,the contents of GSH and GSH-PX in kidneys increased in mice treated with TFS.CONCLUSION Our findings suggested that protective effects of TFS on KBrO3-induced kidney damage related to its free radical scavenging activity and oxidative stress inhibitory effect.
