增殖细胞核抗原与卵巢癌细胞系化学敏感性的关系

摘    要:目的 探讨增殖细胞核抗原 (PCNA)与上皮性卵巢癌细胞系化学敏感性的关系。方法 使用顺铂作用于A 2 780及AD 60细胞株。在MTT法测定耐药倍数基础上 ,用ABC检测PCNA表达。结果 MTT测定AD 60的耐药倍数为 2 .4。A 2 780PCNA的表达低于AD 60 ,且随药物浓度的递增染色较耐药组明显变浅。A 2 780、AD 60相同药物浓度作用后PCNA的表达有显著差异 (P <0 .0 1) ;同一细胞株不同药物浓度作用后PCNA表达有显著性差异 (P <0 .0 1)。结论 增殖细胞核抗原表达与上皮性卵巢癌细胞对细胞毒性药物的化学敏感性呈负相关。

Abstract:Objective To evaluate the relationship between PCNA expression and chemosensitivity in epithelial ovarian cancer cell lines.Method The human epithelial ovarian cancer drug_sensitive cell lines A 2780 and drug_resistance cell lines AD 60 was treated with cisplatin.MTT was used to assay the multiple drug resistance.PCNA expression was studied by avidin_biotin_peroxi_dase complex method(ABC).Results the multiple drug resistance of AD 60 was 2.4.PCNA expression became lighter with the increasing cisplatin concentrations in A 2780 than those in AD 60.there were significant difference of PCNA expression between the two cell lines in the same cisplatin concentrations.Meanwhile,there was significant difference of PCNA expression in the same cell lines in the different cisplatin concentrations.Conclusion The current study suggests that PCNA expression should be related to the chemosensitivity in epithelial ovarian cancer.the higher telomerase activation and PCNA expression,the lower chemosensitivity in cancer cells.
