滤泡调节性T细胞在肺癌移植瘤小鼠体内的变化及意义

摘    要:[摘要]目的: 研究滤泡调节性T细胞(follicular regulatory T cells,Tfr)在肺癌移植瘤小鼠体内的变化,并探讨肿瘤微环境对Tfr细胞的影响。方法: 采用皮下注射Lewis肺癌细胞株的方式建立肺癌移植瘤小鼠模型,通过流式细胞术检测小鼠腹壁引流淋巴结和肿瘤组织中Tfr细胞以及CD4+CXCR5+T细胞的比例,分析Tfr/CD4+CXCR5+T细胞比值的变化。 结果: 肺癌移植瘤小鼠腹壁引流淋巴结中Tfr细胞的比例为(4.13±0.35)%,较正常对照组小鼠[(1.97±0.26)%]明显增加(t=4.985,P<0.01),并且Tfr/CD4+CXCR5+T细胞比值为0.39±0.03,较正常对照组小鼠(0.21±0.02)明显增加(t=5.148,P<0.01)。在肿瘤发展的第14天,肺癌移植瘤小鼠肿瘤组织中Tfr细胞比例增加尤为显著,与第7天相比差异具有统计学意义(t=5.617,P<0.01)。结论: 肺癌移植瘤小鼠体内Tfr细胞比例的增加以及Tfr/CD4+CXCR5+ T细胞比值的失衡,预示着Tfr细胞可能参与小鼠肺癌移植瘤的发展进程。

Abstract:[Abstract]Objective To investigate the change and potential significance of follicular regulatory T cells (Tfr) percentage in murine model of Lewis lung carcinoma mice. Methods The mice were injected with the Lewis cells to establish murine model of lung carcinoma. The percentage of Tfr cells and CD4+CXCR5+T cells in abdominal drainage lymph nodes and tumor tissues were determined by flow cytometer.The change at ratio of Tfr/CD4+CXCR5+T cell were analyzed. Results Compared with the normal control mice (1.97±0.26)%,tumor burdened mice (4.13±0.35)% had a significantly higher percentage of Tfr cells in abdominal drainage lymph nodes(t=4.985, P<0.01). Tfr/CD4+CXCR5+T cell ratio in tumor burdened mice(0.39±0.03) was also obviously increased(t=5.148, P<0.01) compared with the normal control mice(0.21±0.02). The percentage of Tfr cells in tumor tissues had a significantly higher on fourteenth day,the difference was statistically significant compared with seventh days(t=5.617, P<0.01). Conclusion The increased percentage of Tfr cells in Lewis lung carcinoma mice and the imbalance of Tfr/CD4+CXCR5+T cell ratio indicated that Tfr cells may be involved in the progression of lung carcinoma.
