柴胡总皂甙对对戊四氮慢性点燃大鼠海马 GFAP表达的影响

摘    要:目的 研究柴胡总皂甙对戊四氮(PTZ)慢性点燃癫痫模型大鼠海马区星形胶质细胞的胶质纤维酸性蛋白(GFAP)表达的影响。方法48只健康SD大鼠被随机均分为6组,即空白组(A组)、生理盐水组(B组)、丙戊酸钠(VPA)组(C组)和柴胡总皂甙高、中、低3种剂量组(D组、E组、F组),除A组不做处理外,其他各组采用腹腔注射盯z慢性点燃造模,造模同时给予VPA、柴胡总皂甙等不同处理因素,连续4周后取脑组织切片进行免疫组化染色,从阳性细胞数、截面积、灰度值分析结果。结果B组海马各区的阳性细胞数、阳性细胞截面积高于其他各组(P〈0.01)。B组海马各区阳性细胞灰度值低于其他各组,经统计分析CAI区与A组、C组、D组差异显著(P〈01),CA2区与A组、C组、D组、E组差异显著(P〈.05),而在DG区与F组差异有统计学意义(P〈0.05),与A组、C组、D组、E组有显著性差异(P〈0.01)。结论柴胡总皂甙可以抑制PTZ点燃大鼠海马GFAP的过度表达.柴胡总皂甙存在抑制点燃大鼠海马星形胶质细胞的异常激活的作用。

Effect of saikosaponins on glial fibrillary acidic protein expression in hippocampal astrocytes of pentetrazole-induced chronic kindling rats

Abstract:Objective To study the effect of saikosaponins, the active ingredients of Bupleurum chinense DC, on glial fibrillary acidic protein (GFAP) expression in hippocampal astrocytes of chronic kindling rats induced by pentetrazole (PTZ). Methods Forty-eight healthy Sprague-Dawley rats were randomized into 6 equal groups, namely the blank control group (Group A), normal saline group (Group B), sodium valproate group (Group C), and 3 saikosaponins groups of high, medium and small doses (Groups D, E, and F, respectively). The rats (except those in Group A) received intraperitoneal injection of PTZ to induce chronic kindling 1 h after the respective agents as indicated were administered intragastrically on a daily basis for 4 consecutive weeks. Upon completion of the treatment course, the rats were sacrificed and the brain tissues were sampled, sliced and stained for immunohistochemical examination. The results were alalyzed to calculate the positive cell count, cross-sectional area of the cells and the gray scale. Results In group B, the positive cell population and cross-sectional area of the positive cells were the greatest among the groups (P<0.01), but the positive cell gray scale of the CA1 and CA2 regions and the dentate gyrus (DG) of the hippocampus was the lowest. The CA1 region of Group B was significantly different from that of groups A, C and D (P<0.01), and the CA2 region different from groups A, C, D and E (P<0.05), while the DG different from group F (P<0.05) and groups A, C, D and E (P<0.01). Conclusion In chronic kindling rats induced by PTZ, GFAP overexpression can be inhibited by saikosaponins, which suppress the abnormal activation of hippocampal astrocyte of the kindling rats.
