摘    要:目的:对重组克隆表达抗原血清学分析(SEREX)法筛选出的鼻咽癌(NPC)系列肿瘤抗原进行特异性检测,判断新检测出的NPC抗原的相应抗体在NPC病人血清及正常人血清中的阳性率,为NPC抗原对NPC的免疫诊断、免疫治疗积累基础资料。方法:运用噬菌体转染-蛋白印迹、免疫组织化学技术,检测3种NPC肿瘤抗原(GX-NPC-1,GX-NPC-2,GX-NPC-4)在20个NPC病人及15个正常人血清中的阳性表达率。结果:3种NPC肿瘤抗原在NPC病人血清中的阳性表达率分别为55%(11/20)、55%(11/20)、50%(10/20);正常人各为33.3%(5/15)、26.7%(4/15)、6.7%(1/15),经统计学分析,前两种肿瘤抗原在NPC及正常人血清中的表达差异无统计学意义,而GX-NPC-4在两种血清中的表达差异有统计学意义(P<0.01)。结论:3种NPC肿瘤抗原中GX-NPC-4抗原在NPC患者的血清中的表达率明显增高,可能为NPC特异性抗原,值得进一步研究。

Abstract:Objective:The specificity and expression rate of nasopharyngeal carcinoma (NPC) antigens that derived from the SEREX technique (Serological identification of antigens by recombinant cDNA expression cloning) were tested by NPC sera and normal control in order to accumulate information for NPC diagnosis and immunotherapy.Methods:The NPC antigen clones (phage) were transferred into Eacherichia coli. The bacteria were grown in colonies and the lytic plague were transferred by blotting to a nitrocellulose membrane. The diluted human serum was then incubated with the membrane and those antibodies that bind to NPC antigen expressed in lytic plaques were made visible by enzyme-conjugated goat anti-human IgG antibody.Result:Three NPC antigens (GX-NPC-1, GX-NPC-2, GX-NPC-4) reaction rate were 55% (11/20), 55% (11/20), 50% (10/20) in NPC sera, and 33.3% (5/15), 26.7% (4/15), 6.7% (1/15) in normal control, respectively. A chi square test was shown that reaction rate of two antigens (GX-NPC-1 and GX-NPC-2) between NPC sera and normal control was nonsignificant sense. However, the reaction rate of antigen (GX-NPC-4) in NPC sera was significantly higher than that in normal control (P<0.01).Conclusion:The antigen (GX-NPC-4) reaction rate in NPC sera was significantly higher than that in normal control demonstrated that it may be a NPC specific antigen. To explain its biological function and clinical availability, further research of more NPC case and others tumor are necessary.
