| 摘 要： | 目的利用RNAi使解聚素-金属蛋白酶17(ADAM17)基因沉默,观察AD-AM17 mRNA在人MCF-7乳腺癌细胞中的表达及其对细胞增殖的影响。方法利用脂质体介导ADAM17-siRNA转染MCF-7细胞,采用实时荧光聚合酶链反应(Real-time PCR)检测ADAM17 mRNA表达的水平;MTT法检测细胞增殖的活性;流式细胞仪检测细胞周期。结果 ADAM17 mRNA在MCF-7细胞中高表达,转染特异性ADAM17-siRNA后,其表达显著被抑制,同时MCF-7细胞增殖能力也明显下降,细胞进入S和G2/M期的比例降低,绝大多数细胞停留在G0/G1期。结论 ADAM17-siRNA可通过脂质体有效转染MCF-7人乳腺癌细胞,转染后的MCF-7细胞增殖能力明显下降,这种作用与阻止癌细胞进入增殖周期有关。
