猪肺动脉内皮细胞培养液对离体大鼠心脏的影响

摘    要:为了探讨心肌再灌注损伤时,内皮细胞的损伤与心肌损伤之间的因果关系。本实验收集经缺氧再给氧处理的培养猪肺动脉内皮细胞培养液灌流Langendorff大鼠心脏(实验组),观察其对心脏功能与生化指标的影响,并设立相应的对照组。在20—30min的观察期内,与对照组比较实验组的冠脉灌注压(CPP),冠脉流出液中磷酸肌酸激酶(CPK)与乳酸脱氢梅(LDH)显著升高;心肌组织中超氧化物歧化酶(SOD)与ATPase活性明显下降,丙二醛(MDA)水平略高,心率(HR)减慢,心律失常,室内压最大上升速率( dp/dt_(max))降低;室内压最大下降速率(-dp/dtmax)与室内压(LVSP)无明显差异。结果表明:经缺氧再给氧处理的培养猪肺动脉内皮细胞培养液可引起离体大鼠心脏损伤,因此提示,心脏内皮细胞的损伤可能是心肌再灌注损伤的起因。

Effects of the Medium of the Cultured Endothelial Cells from Porcine Pulmonary Artery on the Isolated Rat Hearts

Abstract:In order to research causal relation between endothelial cell damage and myocar-dial injury, effects of the medium of the cultured endotheJial cells, which underwent treatment of re-oxygenation following anoxia (the experimental group), from porcine pulmonary artery on cardie function and biochemical index of Langendorff rat hearts were measured and were compared with those in the control group relating to the experimental group. In the experimental group during the observation time for 20-30 minutes, coronary perfusion pressure, lactate dehy-drogenase and creatine phosphokinase in the coronary excretion fluid significantly increased (activity of superoxide dismutase and ATPase in the myocareial tissue obviously decreased; malonal-de-hyde slightly increased; heart rate reduced; heart rhythm changed (positive maximal rate in ventricular pressure development reduced remarkedly; Left ventricular systolic pressure were not ob-vionsly different, compared with control group. The results suggested that the culture fluid of endothelial cells underwent re-oxygenation following anoxia might cause myocardial damage in the isolated rat hearts. According to this results, we might deduced endothelial cells injury might was one origin about myocardial damage during ischmia and reperfusion.
