大鼠舌癌变过程中MMP-2及MMP-9基因表达的变化

目的　通过观察基质金属蛋白酶 (MMP)在大鼠舌癌变过程中的不同表达 ,分析MMP在口腔鳞癌 (OSCC)及其癌变过程中的表达规律 ,探讨其与肿瘤发生、发展的关系。方法　用原位杂交方法检测 80例 4NQO饮水诱发大鼠舌癌变各期组织MMP 2和MMP 9基因表达。结果　正常粘膜、轻中度异常增生、原位癌、鳞癌的MMP 2mRNA阳性率分别为 8 3% ,2 2 6 % ,35 7% ,73 9% ;MMP 9mRNA阳性率分别为 8 3% ,19 4 % ,35 7% ,82 6 %。结论　MMP 2、MMP 9基因上调表达与口腔粘膜基底膜破坏密切相关 ,是口腔癌浸润生长的重要分子机制。

Expressions of Matrix Metalloproteinase-2 and-9 in Rat Tongue Carcinogenesis

Objective To observe the different expressions of the MMP 2, MMP 9 in oral carcinogenesis, analyze the exhibition rule of MMP 2, 9 OSCC and the process of carcinogenesis, and try to investigate the relationship between the abnormal show of MMP 2, 9, the occurrence and development of carcinoma.Methods A total of 80 rat tongue carcinogenic specimens induced by 4NQO were detected for MMP 2 and MMP 9 gene in situ hybridization.Results With the progress of tongue carcinogenesis and epithelium cell dissifferentiation, the expression of MMP 2, 9 was up regulated. The positive rate of MMP 2 in normal, dysplasia, in situ carcinoma and squamous cell carcinoma was 8 3%,22 6%,35 7% and 73 9%, respectively, while that of MMP 9 was 8 3%,19 4%,35 7% and 82 6%,respectively.Conclusion The up regulation of MMP 2, 9 may be associated with the damage of oral mucoss and may be an important molecular mechanism of oral carcinoma invasion.
