蛋白激酶C-alpha和beta在血压及肾小球滤过功能中的作用比较

摘    要:目的明确蛋白激酶C(PKC)-alpha和beta在小鼠血压及肾小球滤过功能中的不同作用。方法使用代谢箱收集正常饮食下PKC—alpha和beta两种基因敲击鼠及相应野生鼠24h尿液,采用间接免疫酶联吸附法(ELISA)检测血清及24h尿白蛋白排泄情况,3[H]标记的菊粉作清除率试验并观察小鼠血压。结果与野生鼠相比,正常饮食下PKC—alpha基因敲击鼠血压、尿白蛋白正常,肾小球滤过率(GFR)减少(P=0.02),而PKC—beta基因敲击鼠尿白蛋白、GFR正常,血压上升(P=0.03)。结论PKC—alpha主要参与调节肾小球的滤过功能,而PKC—beta主要参与调节动脉血压,每一亚基的具体作用不能被另一亚基取代。

Abstract:Objective To compare the role of protein kinase C(PKC) alpha and beta on blood pressure and glomerular filtration function of mouse.Methods PKC alpha,beta knockout and each pointed wild type mice(n=6,each group)were placed in metabolic cages to assess 24 hours urinary albumin excretion by ELISA.Glomerular filtration rate(GFR) of these mice were determined by clearance experiment performed with labeled inulin and at the same time checked systolic blood pressure by pressure transducer.Results PKC alpha and beta knockout mice exhibited normal 24 hours urinary albumin excretion under basal condition as compared to pointed wild type mice.However,as compared to wild type mice,PKC alpha knockout mice presented normal blood pressure,lower GFR(P=0.02)and PKC beta knockout mice showed high blood pressure(P=0.03)and normal GFR.Conclusion PKC alpha contributes more to mediate GFR,PKC beta plays a major role in regulation arterial blood pressure,and these functions could not be compensated by each other.
